摘要
Objective: To investigate the content of miR-19a in hepatocellular carcinoma (HCC) and its role on the biological function by targeting PTEN. @@@ Methods: miR-19a in 85 patients with liver carcinoma was tested by qPCR, and the content of miR-19a in HCC cells (HepG2, SMMC7721, BEL-7402, Hep3B) and human healthy liver cells (HL-7702) were tested to select suitable experimental cells. The role of miR-19a on the proliferation of HCC cells was determined using CCK-8 and clone formation test. Transwell test and WB test were applied to test the invasion of HCC cells and the content of related invasion proteins. The correlation of miR-19a with PTEN in HCC was detected using luciferase and Western blot. Results: miR-19a was enhanced in HCC. CCK-8 and clone formation test revealed that the proliferation of SMMC7721 transfected with miR-19a inhibitor was obviously weakened. Transwell suggested that decrease of miR-19a in HCC cells could weaken the invasion of HCC cells, and E-cadherin protein enhanced evidently, while Vimentin protein reduced. Flow cytometry suggested that the apoptosis of miR-19a inhibitor enhanced evidently. Double fluorescein report suggested that PTEN was the target of miR-19a. Conclusion: miR-19a may affect the proliferation, invasion, and apoptosis of HCC cells by elevating PTEN.
