摘要

The present study investigated the effects of normal and reduced changeable ambient UV-B irradiation on the accumulation of diterpene lactones and flavonoids in Andrographis paniculata (Burm.f.) Nees. The plants were respectively cultured under the conditions of ambient UV-B irradiation (UV-B-1), slightly low ambient UV-B irradiation (UV-B-2) and severely low ambient UV-B irradiation (UV-B-3) for 28 days, then sampled at the 7th, 14th, 21st and 28th day respectively to determine the two diterpene lactones (andrographolide (A); deoxyandrographolide (DA)), two flavonoids (7-O-methyldihydrowogonin (F1); 7-O-methylwogonin (F2)), total flavonoids and total lactones. The results indicated that lowering ambient UV-B irradiation was not beneficial for the accumulations of total lactones, total flavonoids, DA, F1 and F2, but was good for the increment of A in Andrographis paniculata. Compared with UV-B-1 condition, the contents of total lactones, total flavonoids, DA, F1, F2 and A in Andrographis paniculata decreased respectively by 1.84, 29.40, 47.52, 49.21, 10.54 and 27.42% under UV-B-2 condition, or decreased respectively by 3.12, 0.83, 35.30, 38.11, 18.58 and 12.14% under UV-B-3 condition at the end of 7-day treatments. But the changes of their contents were out of proportion to the descending intensity of UV-B irradiation. The severely low ambient UV-B irradiation (UV-B-3) could only decrease the contents of A and F2 a little (decreased by 11.99, 8.98%, correspondingly) or increased the contents of DA and F1 (increased by 23.29, 21.85%, correspondingly) in the plant by contrast with the slightly low ambient UV-B irradiation (UV-B-2). During the 28-day experiment, the intensity of ambient UV-B irradiation was changeable with the trend of sharp decrease - slight decrease - slight increase. Whether in the normal or in the reduced ambient UV-B irradiation, the accumulations of total lactone and DA in Andrographis paniculata was consistent with the variation of ambient UV-B irradiation, while the contents of total flavonoid and A slowly decreased all the time, and the accumulation of two flavonoids (F1 and F2) showed a slight increase-sharp increase-sharp decrease trend at that period. This study indicated the promoting or inhibiting effects of ambient UV-B irradiation on the accumulation of different diterpene lactones or flavonoids in Andrographis paniculata varied from constituents, and it was very important to select suitable intensity of ambient UV-B irradiation to promote the accumulation of active constituents during the plant cultivation.
