Arthritis biosensing: Aptamer-antibody-mediated identification of biomarkers by ELISA

摘要

Arthritis is a common disease in joints, causing tenderness, swelling and joint pain. Early identification of arthritis helps to prevent joint erosion, slow erosive disease progression, and achieve remission after treatment. Interleukin-6 (IL-6) is a cytokine biomarker that is overexpressed in response to inflammation, injury, and infection. Since elevation of IL-6 was found by researchers with arthritis patients, quantification of IL-6 helps for the early identification of arthritis. In this research, high affinity dual probed ELISA was introduced to identify IL-6 at low levels. On the gold nanourchin, the antibody and aptamer were attached and used as the detection probes to identify IL-6 on the ELISA surface. The dual probe was found to be highly sensitive and selective to IL-6 and bound more IL-6. The detection limit of IL-6 with the dual probe was calculated as 0.5 pg/mL by linear regression with an R-2 value of 0.9889. Furthermore, control experiments without detection probes or target and with the control protein IL-6 receptor did not show significant absorbance changes, confirming the specific detection of IL-6. This ELISA method with a dual probe identifies a lower abundance of IL-6 and helps to diagnose arthritis.
