Degradation can be identified due to the presence of a downward slope across the electropherogram, where larger loci have a lower probability to be amplified and thus a major chance of dropping out. The introduction of the degradation index (DI) of Quantifiler® Trio and the optimization of the available amplification kits have been improving the genotyping success of these samples. Casework samples were amplified with GlobalFiler™ and PowerPlex® Fusion 6C, according to the quantification results. Quantification and amplification results demonstrated that it is important to consider DI and DNA concentration values to define downstream procedure and also that the genetic information provided by both amplification kits could be useful for degraded samples profiling.
