A new green microalga was isolated from a salt lake in Tibet Plateau, China, classified as genus Desmodesmus under the family of Scenedesmaceae based on morphological and 18S rDNA sequencing. The species was able to grow at both 4 and 25 degrees C, due to differences to the biochemical composition. At 25 degrees C, protein content made up 50 % in dry biomass, while at 4 degrees C lipids made up 55 % of dry biomass. A poly unsaturated fatty acid, cis-8,11,14- docosatrienoic acid (C22:3, 8,11,14-DTA), was identified in D. sp. QL96 cultured under both temperature con-ditions. This fatty acid has been reported to be physiologically functional, however yield from natural sources was low and limited the related research and application. GC analysis indicated that 8,11,14-DTA was the third abundant fatty acid in D. sp. QL96, accounting for around 20 % out of the total lipid. The effects of environ-mental factors on the production of 8,11,14-DTA was studied and productivity of 8,11,14-DTA was optimized to achieve 10.91 mg/L per day. To our knowledge, the plateau alga is the most productive natural source of 8,11,14-DTA. This study provides the groundwork for further study and processing of this fatty acid.
