DIOSCOREA SEPTEMLOBA THUNB GRANULES DECREASES SERUM URIC ACID LEVEL IN HYPERURICEMIA RAT VIA REGULATING OATP1A1 AND OCTN2 LEVELS

Summary

Purpose: The purpose of this study was to explore the effect of DST granules on the serum uric acid (UA) level in HUA rats. @@@ Methods: The rat hyperuricemia (HUA) model was established by administrating adenine (200 mg/ kg/ d) for 21 days. The HUA rats were given DST at 120 mg/ kg/ d (low-dose group), 240 mg/ kg/ d (mild-dose group), or 480 mg/ kg/ d (high-dose group) for 14 days by oral administration. At the same time, normal rats and HUA rats were given equivalent 15% CMC-Na. The mRNA and protein expression levels of ATPase Na+/ K+ transporting subunit alpha 1 (ATP1a1) and organic cation/ carnitine transporter 2 (OCTN2) were evaluated by PCR, Western blot, and immunohistochemical staining. @@@ Results: DST significantly decreased the HUA-induced elevation of serum UA, Cr, and BUN levels (P <0.05). The serum UA level was decreased by DST in a dose-dependent manner (P <0.05). DST clearly attenuated HUA induced decreased levels of OATP1a1 and OCTN2 in the liver, kidney, stomach, and small intestine tissues in a dose-dependent manner (P <0.05). @@@ Conclusion: DST may decrease serum levels of UA, Cr, and BUN through up- regulation of OATP1a1 and OCTN2 in the liver, kidney, stomach, and small intestine tissues.
